Biological Background: PI3 Kinase p85 alpha Function and Localization
- Phosphatidylinositol 3-kinase regulatory subunit alpha (PI3K p85 alpha) is the 85 kDa regulatory subunit of class IA PI3Ks, encoded by the PIK3R1 gene. It is also known as p85, GRB1, and p85alpha.
- Acts as an adapter protein that binds to activated phosphorylated protein-tyrosine kinases via its SH2 domain, mediating the association of the p110 catalytic subunit to the plasma membrane. This interaction is necessary for insulin-stimulated glucose uptake and glycogen synthesis, and plays a role in signaling through FGFR, KIT, PDGFRA/B, and ITGB2. - Modulates the cellular response to ER stress by promoting nuclear translocation of XBP1 isoform 2 in an ER stress- and/or insulin-dependent manner, contributing to improved glucose tolerance in the liver. Related references: PMID:20348923

- Isoform 2 is expressed in skeletal muscle and brain, with lower levels in kidney and cardiac muscle; isoforms 2 and 4 are detected at the protein level in skeletal muscle.
