Biological Background: Phospho-β-Catenin (Ser552) Function and Localization

  • Catenin beta-1 (β-catenin) is a dual-function protein acting both in cell adhesion and in Wnt signal transduction. It links E-cadherin to the actin cytoskeleton at adherens junctions and shuttles to the nucleus to activate TCF/LEF target genes.

  • In the absence of Wnt ligand, β-catenin is phosphorylated on N-terminal Ser/Thr residues, ubiquitinated by the β-TrCP E3 ligase, and degraded by the proteasome. Upon Wnt stimulation, β-catenin is stabilized and translocates to the nucleus (PubMed:17524503, PubMed:18077326, PubMed:18086858).
