Biological Background: ULK1 Function and Localization
- Serine/threonine-protein kinase involved in autophagy in response to starvation. Acts upstream of phosphatidylinositol 3-kinase PIK3C3 to regulate the formation of autophagophores. Part of regulatory feedback loops with mTORC1 and AMPK. Related references: PMID:18936157 PMID:21460634 PMID:21795849
- The ULK1 gene encodes the human homolog of yeast ATG1, a key autophagy initiator, and is a member of the unc-51-like kinase family.
- Localizes to the cytoplasm, cytosol, and preautophagosomal structures, consistent with its role in early autophagosome formation.
- Activated via phosphorylation by AMPK and in turn regulates AMPK activity by phosphorylating its subunits PRKAA1, PRKAB2, and PRKAG1. Related references: PMID:21460634
- Phosphorylates multiple autophagy-related substrates including AMBRA1, FLCN, SESN2, SQSTM1, and ATG4B, thereby regulating autophagosome nucleation, cargo recruitment, and ATG4B activity. Related references: PMID:20921139 PMID:25126726 PMID:25040165 PMID:37306101 PMID:28821708
- Plays a role early in neuronal differentiation and is required for granule cell axon formation. Related references: PMID:11146101
- Ubiquitously expressed, with detection in adult skeletal muscle, heart, pancreas, brain, placenta, liver, kidney, and lung.
- Annotated with keywords: Acetylation, ATP-binding, Autophagy, Cytoplasm, Kinase, Nucleotide-binding, Phosphoprotein, Ubl conjugation.
Experimental Guidance and Technical Tips
- This antibody is designed to specifically detect phosphorylated ULK1 at Ser555. For optimal Western blot results, consider including phosphatase inhibitors in lysis buffers to preserve phosphorylation status.
- Validate the antibody in the relevant sample system under endogenous conditions; phospho-specific signals may require confirmation via phosphatase treatment or blocking peptide competition experiments.
- The immunogen is derived from a human sequence; cross-reactivity with other species may be limited. Please validate if using non-human samples.
- For ELISA applications, pairing with a total ULK1 antibody may allow normalization to total protein levels.
