Biological Background: Microtubule-associated protein tau Function and Localization

  • Microtubule-associated protein tau (MAPT) is a neuronal phosphoprotein that promotes microtubule assembly and stability, and plays a critical role in the establishment and maintenance of neuronal polarity. It acts as a linker between axonal microtubules and the plasma membrane. Related references: PMID:21985311 PMID:32961270
  • Alternative splicing of the MAPT gene generates multiple isoforms; short isoforms confer cytoskeletal plasticity, while longer isoforms predominantly stabilize microtubules.
  • Tau is predominantly expressed in neurons of the central nervous system, while the PNS-tau isoform is specific to the peripheral nervous system.
  • Subcellularly, tau localizes to the cytoplasm, cytosol, cytoskeleton, and cell membrane, with enrichment in axons and dendrites; it is also secreted.
  • Tau undergoes extensive post-translational modifications including phosphorylation (e.g., at T181, S202, T231), acetylation, methylation, glycation, and ubiquitination, many of which are implicated in tauopathies.
  • Pathological hyperphosphorylation and aggregation of tau lead to neurofibrillary tangles, a hallmark of Alzheimer disease and related neurodegenerative disorders; mutations in MAPT cause frontotemporal dementia (FTDP-17).
  • The dynamic balance between microtubule-bound and free tau regulates neurite outgrowth, axonal transport, and synaptic plasticity.

Experimental Guidance and Technical Tips

  • Due to the phosphorylation-specific nature of this antibody, confirm specificity by treating cell lysates with lambda protein phosphatase, which should abolish signal in Western blot.
  • For immunohistochemical applications, consider antigen retrieval conditions carefully; phospho-epitopes may be sensitive to heat and pH.
  • Include both total Tau and loading controls to assess phosphorylation changes accurately.
  • When detecting endogenous Tau, note that the protein runs at an apparent molecular weight of 45–68 kDa on SDS-PAGE, with the calculated weight of the longest isoform being 78.9 kDa; multiple bands may represent splice variants and post-translational modifications.
  • For co-immunoprecipitation, use fresh samples with phosphatase inhibitors to preserve the phosphorylation state.
