Biological Background: Microtubule-associated Protein Tau Function and Localization
- Microtubule-associated protein tau (MAPT) is a neuronal phosphoprotein that stabilizes and promotes the assembly of microtubules, particularly in axons, and acts as a linker between microtubules and the plasma membrane through its C-terminus and N-terminus, respectively. - Multiple tau isoforms are generated by alternative splicing, with shorter isoforms allowing cytoskeletal plasticity and longer isoforms preferentially stabilizing microtubules in the central nervous system.
- Tau localizes to the cytoplasm, cytosol, cytoskeleton, axons, dendrites, and cell membrane, and can also be secreted extracellularly.
- Expressed predominantly in neurons, with isoform PNS-tau specific to the peripheral nervous system and other isoforms in the central nervous system.
