Biological Background: STK11 Function and Localization

  • STK11 (also known as LKB1, hLKB1, or PJS) is a tumor suppressor serine/threonine-protein kinase that regulates cell metabolism, polarity, apoptosis, and DNA damage response.
  • Acts as a master upstream kinase for the AMPK family, phosphorylating their T-loops to promote activity and thereby controlling energy homeostasis and cell growth (PubMed:12805220, PubMed:14517248, PubMed:14976552).
    Related references: PMID:12805220 PMID:14517248 PMID:14976552
  • Phosphorylates a wide range of substrates including PRKAA1, PRKAA2, BRSK1, BRSK2, MARK1-4, NUAK1/2, SIK1-3, and SNRK, but not MELK (PubMed:12805220, PubMed:14517248, PubMed:14976552).
    Related references: PMID:12805220 PMID:14517248 PMID:14976552
  • Located in multiple subcellular compartments including the nucleus, cytoplasm, membrane, mitochondrion, and late endosomes, consistent with its diverse biological functions.
  • Expressed ubiquitously, with strongest expression in testis and fetal liver; also plays a role in spermiogenesis.
  • Regulated by various post-translational modifications (e.g., acetylation, methylation, palmitoylation, prenylation, phosphorylation). Phosphorylation at S428 (the epitope recognized by this antibody) is implicated in modulating STK11 activity and interactions.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic phosphopeptide around S428; therefore, this antibody is expected to specifically recognize STK11 only when phosphorylated at S428. Non-phosphorylated STK11 may not be detected.
  • For Western blotting, include phosphatase inhibitors in the lysis buffer to preserve phosphorylation signals. Consider using positive controls such as cells treated with stimuli known to induce STK11 phosphorylation at S428.
  • In ELISA, the antibody may be used to detect phospho-peptide antigens; optimization of coating conditions and antibody dilution is recommended.
  • Always validate the antibody in the relevant sample system and under your specific experimental conditions to confirm specificity and performance.
