Biological Background: STING/TMEM173 Function and Localization

  • Stimulator of interferon genes protein (STING/TMEM173), also referred to as ERIS, MITA, MPYS, and hSTING, is encoded by the STING1 gene in humans and exists as multiple isoforms including STING-beta.

Experimental Guidance and Technical Tips

  • Since the immunogen is a synthetic phosphorylated peptide around Ser366, this antibody is expected to specifically recognize STING/TMEM173 only when phosphorylated at this site, which occurs upon activation of the cGAS-STING pathway (e.g., after dsDNA or cGAMP stimulation).
  • For Western blotting (WB), phosphorylated STING can be detected in human cell lysates following treatment with cGAMP or dsDNA to stimulate the pathway; a band at approximately 42 kDa is expected. Non-phosphorylated STING should not be detected.
  • In ELISA applications, this antibody can be used to capture or detect phosphorylated STING in cell or tissue extracts; consider pairing with a total STING antibody for normalization or sandwich assays.
