Biological Background: STING Function and Localization
- STING (stimulator of interferon genes, also known as TMEM173/ERIS/MITA) is a facilitator of innate immune signaling that senses cytosolic DNA from bacteria and viruses and promotes the production of type I interferons (IFN-α and IFN-β). - It acts by binding cyclic dinucleotides including bacterial cyclic di-GMP (c-di-GMP), and endogenous cyclic GMP-AMP (cGAMP) produced by cGAS upon cytosolic DNA recognition. Related references: PMID:21947006, PMID:23258412, PMID:23519410
- Upon ligand binding, STING oligomerizes, translocates from the endoplasmic reticulum to the Golgi, and is phosphorylated by TBK1 at the pLxIS motif (including S365), leading to IRF3 recruitment and interferon induction. Related references: PMID:25636800, PMID:27324217, PMID:29973723
- STING also triggers autophagy, with cGAMP binding inducing its movement to ERGIC and autophagosome formation to degrade cytosolic DNA pathogens. The autophagy function can be uncoupled from interferon signaling. Related references: PMID:30568238
- STING is ubiquitously expressed but enriched in immune tissues such as spleen and thymus, and in dendritic cells.
- Subcellularly, STING resides on the endoplasmic reticulum membrane, and upon activation traffics through the ERGIC and Golgi, and can localize to autophagosomes, mitochondria, and the cell membrane.
- Phosphorylation at serine 365 (S365) within the pLxIS motif is critical for STING-mediated IRF3 activation and interferon production. The S365 phosphorylated form is a key activation marker.
