Biological Background: STAT3 Function and Localization
- STAT3, also known as acute-phase response factor, is a signal transducer and transcription activator that mediates cellular responses to interleukins, KITLG/SCF, LEP, and other growth factors (PubMed:10688651, PubMed:11294841, PubMed:12359225). Upon activation, it recruits coactivators such as NCOA1 or MED1 to target gene promoters (PubMed:15653507, PubMed:16285960, PubMed:17344214). It binds IL6-responsive elements in acute-phase protein gene promoters (PubMed:12359225) and is activated by IL31 through IL31RA (PubMed:15194700). Related references: PMID:10688651 PMID:11294841 PMID:12359225 PMID:15653507 PMID:16285960 PMID:17344214 PMID:15194700
- STAT3 regulates inflammatory responses by controlling differentiation of naive CD4+ T-cells into T-helper Th17 or regulatory T-cells (Treg). Acetylation enhances its transcriptional activity and cell differentiation, while deacetylation and oxidation by LOXL3 inhibit differentiation (PubMed:28065600, PubMed:28262505). Related references: PMID:28065600 PMID:28262505
- Involved in cell cycle regulation by inducing expression of key genes like CCND1 for progression from G1 to S phase (PubMed:17344214). Related references: PMID:17344214
- STAT3 mediates LEP effects on melanocortin production, body energy homeostasis, and lactation (By similarity) and may promote apoptosis via BIRC5 transactivation under LEP stimulation (PubMed:18242580). Related references: PMID:18242580
- Cytoplasmic STAT3 represses macroautophagy by inhibiting EIF2AK2/PKR activity (PubMed:23084476). Related references: PMID:23084476
- Plays a crucial role in basal beta cell functions, such as regulation of insulin secretion (By similarity).
- Involved in oncostatin-M-mediated signaling through both type I and type II OSM receptor complexes (PubMed:9188471). Related references: PMID:9188471
- Expressed in heart, brain, placenta, lung, liver, skeletal muscle, kidney, and pancreas, as well as naive CD4+ T cells and T-helper Th17, Th1, Th2 cells (PubMed:31899195). Related references: PMID:31899195
- Subject to multiple post-translational modifications including phosphorylation, acetylation, methylation, and ubiquitination-like conjugation.
- Localizes to both cytoplasm and nucleus.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic phospho-peptide around Tyr705, a key residue phosphorylated by JAK kinases upon cytokine stimulation.
- For WB, consider using phosphatase inhibitors during lysis to maintain phosphorylation signals.
- Validate the antibody in an appropriate cell system, e.g., IL-6-treated HeLa cells, to confirm specificity for phospho-Y705.
- The antibody is suitable for IP and ELISA; optimal working concentrations should be determined empirically.
