Biological Background: SMAD2 Function and Localization
- SMAD2 (Mothers against decapentaplegic homolog 2), also known as JV18-1, MADH2, or hMAD-2, is a receptor-regulated SMAD (R-SMAD) protein that functions as an intracellular signal transducer for TGF-beta and activin pathways.
- Upon ligand binding, SMAD2 is phosphorylated by type 1 receptor kinases, forms a complex with SMAD4, and translocates to the nucleus where it binds the TRE element to regulate transcription of target genes.
- SMAD2 positively regulates PDPK1 kinase activity by promoting its dissociation from the inhibitory 14-3-3 protein YWHAQ.
