Biological Background: Smad1 Function and Localization

  • Smad1 (Mothers against decapentaplegic homolog 1, MADH1, BSP1) is a key intracellular mediator of the bone morphogenetic protein (BMP) signaling pathway, belonging to the SMAD family of transcription factors. It is also known as JV4-1, MADR1, and BSP-1.
  • Functions as a transcriptional modulator regulating embryonic development, cell differentiation, and tissue homeostasis by binding to Smad Binding Elements (SBEs) in target gene promoters (PubMed:9335504, PubMed:33667543). Related references: PMID:9335504 PMID:33667543
  • Upon BMP receptor activation, Smad1 is phosphorylated at the C-terminal SSXS motif (including Ser206) by type I BMP receptors, enabling complex formation with SMAD4 and nuclear translocation to regulate transcription (PubMed:33667543). Related references: PMID:33667543
  • Subcellular localization is both cytoplasmic and nuclear; in the absence of signaling, Smad1 resides mainly in the cytoplasm, and upon activation, it accumulates in the nucleus.
  • Ubiquitously expressed with highest levels in heart and skeletal muscle, reflecting its broad roles in tissue patterning and homeostasis.
  • Post-translational modifications include phosphorylation, acetylation, and ubiquitination, which modulate its stability, activity, and interactions.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic phosphopeptide around Ser206; consider using phosphatase inhibitors during sample preparation to maintain the phosphorylation state.
  • For Western blotting (WB), a band of approximately 52 kDa is expected. Validate the antibody with appropriate positive and negative controls (e.g., untreated vs. BMP-treated lysates).
  • Cross-reactivity is reported for human, mouse, and rat; confirm specificity in your experimental model by siRNA knockdown or peptide competition assays.
  • ELISA applications may require titration to determine the optimal coating concentration and detection range.
