Biological Background: Replication Protein A 32 kDa Subunit Function and Localization

  • The Replication protein A 32 kDa subunit (RPA2, also known as RPA32, RP-A p32, RP-A p34) is a component of the heterotrimeric replication protein A (RPA) complex, which includes RPA1, RPA2, and RPA3.
  • RPA binds and stabilizes single-stranded DNA (ssDNA) intermediates generated during DNA replication and DNA stress, preventing reannealing and facilitating the recruitment of DNA metabolism proteins.
  • In the cellular response to DNA damage, RPA is crucial for DNA repair and checkpoint activation. It recruits the ATR-ATRIP kinase complex to DNA damage sites, initiating the DNA damage response cascade.
  • RPA recruits key repair factors to chromatin, including RAD51 and RAD52 for double-strand break repair, as well as XPA and XPG for nucleotide excision repair (NER). It also participates in base excision repair (BER) via interaction with UNG.

  • The immunogen is a synthetic phosphorylated peptide around S4 and S8 of human RPA2. For Western blot confirmation, consider using phosphatase-treated cell lysates to verify phosphorylation-specific signal loss.
  • The predicted molecular weight of unmodified RPA2 is 29 kDa; however, phosphorylation or other modifications may affect migration. Use appropriate molecular weight markers and validate in your sample system.
  • This antibody is recommended for WB and ELISA. For ELISA, titrate the antibody to determine optimal working dilution, and include a non-phosphorylated peptide control to ensure specificity.
  • Cross-reactivity with rat RPA2 has been reported; if using rat samples, validate performance under your experimental conditions.
