Biological Background: RELB Function and Localization

  • RELB is a member of the NF-kappa-B family of transcription factors, which regulate a broad spectrum of biological processes including inflammation, immunity, cell growth, differentiation, and apoptosis.
  • NF-kappa-B dimers are formed by combinations of Rel-like domain proteins (RELA, RELB, NFKB1, NFKB2, REL); RELB preferentially heterodimerizes with NFKB2/p52 or p50 to act as a transcriptional activator.
  • In resting cells, NF-kappa-B complexes are sequestered in the cytoplasm by I-kappa-B inhibitors; upon stimulation, I-kappa-B phosphorylation and degradation release NF-kappa-B for nuclear translocation and target gene activation.
  • RELB is essential for adaptive immunity, being required for both T and B lymphocyte maturation and function (PubMed:26385063). Related references: PMID:26385063
  • RELB dynamically localizes to the cytoplasm, nucleus, as well as the centrosome and microtubule organizing center.
  • RELB is a phosphoprotein, and phosphorylation at specific residues, including Ser573, modulates its transcriptional activity and protein interactions.
  • Additionally, RELB participates in circadian rhythm regulation by repressing CLOCK-BMAL1 activity and contributes to cancer cell survival through the NUPR1/RELB/IER3 pathway, conferring resistance to stress and chemotherapy.

Experimental Guidance and Technical Tips

  • Because this antibody is phospho-specific, always include phosphatase inhibitors in lysis buffers and confirm specificity by phosphatase treatment or peptide competition assays.
  • For Western blot, optimize antibody dilution and protein loading based on your sample type; the expected band is around 62 kDa, though phosphorylation may cause minor mobility shifts.
  • In ELISA applications, this antibody can serve as either a capture or detection reagent; pairing with a total RELB antibody enables quantitative analysis of RELB phosphorylation.
  • Cross-reactivity has been confirmed with mouse; for other species, verify sequence homology around the S573 site before use.
