Biological Background: Retinoblastoma-associated protein Function and Localization
- Retinoblastoma-associated protein (RB1, also known as pRb, p105-Rb, p110-RB1, pp110) is a tumor suppressor protein encoded by the RB1 gene in humans.
- RB1 is a key regulator of the G1/S transition of the cell cycle. In its hypophosphorylated state, it binds and inhibits E2F transcription factors, repressing transcription of genes required for S-phase entry. Related references: PMID:10499802
- Cyclin-dependent kinase (CDK)-mediated phosphorylation of RB1 disrupts its interaction with E2Fs, releasing the transcriptional repression and allowing cell cycle progression. Related references: PMID:10499802
- RB1 also promotes the G0-G1 transition through phosphorylation by CDK3/cyclin-C, further highlighting its regulatory role in cell cycle entry and exit. Related references: PMID:15084261
- Subcellularly, RB1 is predominantly nuclear but can also be found in the cytoplasm, consistent with its functions in transcription regulation and chromatin organization.
- RB1 is involved in heterochromatin formation and epigenetic silencing by recruiting histone methyltransferases (e.g., SUV39H1, KMT5B, KMT5C) and histone deacetylases, thereby maintaining chromatin structure.
- Tissue expression includes the retina and foreskin keratinocytes, where RB1 protein has been detected.
- Additionally, RB1 is targeted by viral oncoproteins such as SV40 large T antigen, HPV E7, and adenovirus E1A, which disrupt RB1-E2F complexes to promote viral replication.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic phosphopeptide corresponding to residues surrounding Ser795 of human RB. When assessing specificity, consider comparing signals with those from a non-phospho-specific RB antibody or phosphatase-treated samples.
- For Western blotting, RB1 is a large protein (approximately 106 kDa); ensure proper transfer conditions and use prestained markers to confirm molecular weight.
- In immunohistochemistry (IHC-P), antigen retrieval and blocking steps may influence staining; optimize these parameters in your specific tissue system.
- The antibody shows cross-reactivity with mouse and rat RB1, enabling its use in studies involving rodent cell lines or tissues; still, validate expression levels and phosphorylation state in your model.
- Given its application in ELISA, ensure that the phosphorylated epitope is preserved in the coating buffer and that blocking agents do not interfere with phospho-specific binding.
