Biological Background: RB1 Function and Localization
- RB1 (Retinoblastoma-associated protein) is a tumor suppressor that controls cell cycle progression by regulating the G1/S transition, primarily through its interaction with E2F transcription factors. Related references: PMID:10499802
- The hypophosphorylated form of RB1 binds and inhibits E2Fs, while CDK-mediated phosphorylation (e.g., at Ser608) disrupts this interaction, allowing E2F-dependent transcription and S-phase entry. Related references: PMID:10499802
- RB1 also promotes G0-G1 transition via phosphorylation by CDK3/cyclin-C. Related references: PMID:15084261
- RB1 recruits histone methyltransferases and maintains heterochromatin structure, particularly H4K20 trimethylation, and mediates transcriptional repression through HDAC recruitment.
- RB1 is expressed in the retina and in foreskin keratinocytes (at protein level), with subcellular localization in both the nucleus and cytoplasm. Related references: PMID:20940255
- Viral oncoproteins such as SV40 large T antigen, HPV E7, and adenovirus E1A bind RB1, disrupting RB1-E2F complexes and contributing to cellular transformation.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic phosphorylated peptide corresponding to Ser608 of human RB1; for optimal detection, consider using protein lysates from cell lines with known RB1 expression and phosphorylation status.
- Phosphorylated proteins are labile; include phosphatase inhibitors in lysis and assay buffers to preserve the phospho-epitope.
- For Western blotting, a reduction step with DTT or β-mercaptoethanol is recommended; validate the antibody in the relevant sample system, as cross-reactivity with non-phosphorylated RB1 may occur.
- For ELISA, direct or sandwich formats may be optimized; pair with a total RB1 antibody to normalize phosphorylation signals.
