Biological Background: Disks Large Homolog 2 (DLG2) Function and Localization
- Disks large homolog 2 (DLG2), also known as postsynaptic density protein PSD-93 or chapsyn-110, is a scaffold protein encoded by the DLG2 gene.
- It is required for the perception of chronic pain through NMDA receptor signaling. DLG2 regulates the surface expression of NMDA receptors in dorsal horn neurons of the spinal cord (by similarity).
- DLG2 interacts with the cytoplasmic tails of NMDA receptor subunits and inward rectifying potassium channels, contributing to synaptic organization.
- As a component of the postsynaptic density at excitatory synapses, it plays a role in maintaining synaptic stability, particularly at cholinergic synapses.
- Subcellularly, DLG2 localizes to the cell membrane, postsynaptic density, synapse, axon, and perikaryon.
- The protein undergoes post-translational modifications including palmitoylation (a lipoprotein modification) and phosphorylation at multiple sites; the Y340 residue is a phospho-acceptor site targeted by this antibody.
- Keywords: SH3 domain, PDZ domain repeat, alternative splicing, phosphoprotein.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic phosphorylated peptide around Y340. To confirm phospho-specificity, compare signals with and without phosphatase treatment or use a non-phosphopeptide blocking control.
- For Western blotting, include phosphatase inhibitors in the lysis buffer to preserve phosphorylation. A band near 98 kDa is expected for DLG2.
- The antibody is reported to react with human, mouse, and rat samples; validate in your specific cell or tissue system considering the expression profile of DLG2.
- For ELISA, the antibody can be used as a capture or detection reagent against the phospho-Y340 peptide; optimize coating and detection conditions.
