Biological Background: POLR2A Function and Localization
- POLR2A encodes the largest subunit of RNA polymerase II, which catalyzes transcription of mRNA precursors and many non-coding RNAs.
- Forms the active center with POLR2B, coordinating Mg2+ ions for nucleotide addition and proofreading via 3'->5' exonuclease activity. Related references: PMID:30190596 PMID:8381534
- Contains a unique C-terminal domain (CTD) with 52 heptapeptide repeats (YSPTSPS) that serves as a platform for factors regulating transcription initiation, elongation, and termination.
- CTD phosphorylation at Ser5 promotes promoter escape, while Ser2 phosphorylation is required for pause release and pre-mRNA processing. Related references: PMID:26124092
- Acetylation and methylation of CTD lysines regulate gene expression, with methylation having a repressive effect.
- Exhibits RNA-dependent RNA polymerase activity, extending non-coding RNAs at the 3' end.
- Subcellular localization includes nucleus, cytoplasm, and chromosome.
- Relevant keywords: phosphoprotein, acetylation, methylation, DNA-directed RNA polymerase, transcription, hydrolase.
Experimental Guidance and Technical Tips
- This phospho-specific antibody recognizes POLR2A only when Ser2 in the CTD repeat is phosphorylated. Include phosphatase inhibitors in lysis and wash buffers to preserve the phosphorylation signal.
- For chromatin immunoprecipitation (ChIP) and CUT&Tag, consider titrating the antibody and optimizing cross-linking/sonication conditions for your specific target loci.
- The antibody is suitable for Western blot (217 kDa), immunofluorescence, and immunohistochemistry; validate in the relevant sample system using appropriate positive and negative controls (e.g., untreated vs. kinase inhibitor-treated cells).
- Due to the high molecular weight, use a low-percentage SDS-PAGE gel or extended transfer times for WB.
