Biological Background: Phospholamban Function and Localization
- Phospholamban (PLN), also known as PLB, is a 52-amino acid transmembrane phosphoprotein predominantly expressed in cardiac muscle.
- It reversibly inhibits the sarcoplasmic reticulum Ca2+-ATPase SERCA2 (ATP2A2) by reducing its apparent affinity for Ca2+, thereby slowing Ca2+ re-uptake during muscle relaxation (PubMed:28890335). Related references: PMID:28890335
- PLN also modulates cardiac contractility in response to β-adrenergic stimulation; phosphorylation at Thr17 by CaMKII relieves SERCA2 inhibition, enhancing Ca2+ transport.
- Subcellularly, PLN localizes to the endoplasmic/sarcoplasmic reticulum membrane and also to the mitochondrial membrane, where it may influence calcium signaling and energy metabolism.
- PLN activity is regulated by its oligomeric state and post-translational modifications, including phosphorylation, acetylation, and palmitoylation, which affect its inhibitory function.
- Mutations in PLN are linked to dilated cardiomyopathy (CMD1P) and hypertrophic cardiomyopathy (CMH18), highlighting its critical role in cardiac function.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic phosphorylated peptide around Thr17; this antibody is expected to detect phospholamban only when phosphorylated at this site. Avoid using this antibody for detecting total PLN.
- For Western blot (WB), note the predicted molecular weight is 6 kDa; consider using a high-percentage gel (e.g., 12–15% Tris-tricine) and careful transfer conditions for small proteins.
- For immunohistochemistry (IHC-P), validate in cardiac muscle tissue where PLN is highly expressed; consider antigen retrieval methods appropriate for phospho-specific antibodies.
- For ELISA, the peptide-based immunogen suggests the antibody may work well in peptide competition assays; validate in the relevant sample system.
- Cross-reactivity is reported for human, mouse, and rat; for other species, check sequence homology around the phosphorylation site.
