Biological Background: Phospho-PLC gamma 1 (PLCG1)-Y783 Function and Localization
- PLCG1 encodes 1-phosphatidylinositol 4,5-bisphosphate phosphodiesterase gamma-1, a key enzyme in phosphoinositide signaling. It hydrolyzes PIP2 to generate the second messengers DAG and IP3, which mobilize calcium and activate protein kinase C.
- It is activated downstream of receptor tyrosine kinases (e.g., PDGFRA, PDGFRB, EGFR, FGFRs), playing a critical role in transducing growth factor signals to intracellular pathways.
- PLCG1 participates in actin reorganization and cell migration, as shown by its requirement for efficient chemotaxis (PubMed:17229814). Related references: PMID:17229814
- The protein localizes to lamellipodia and membrane ruffles, dynamic structures at the leading edge of migrating cells, underscoring its role in cytoskeletal regulation.
- PLCG1 contains SH2 and SH3 domains that mediate interactions with phosphorylated tyrosine residues on activated receptors, and itself is phosphorylated at key tyrosine residues (including Y783) for full activation.
- It is subject to various post-translational modifications, including phosphorylation and acetylation, and is classified as a phosphoprotein, hydrolase, and transducer.
Experimental Guidance and Technical Tips
- This antibody is raised against a phosphorylated peptide; therefore, it specifically recognizes PLCG1 phosphorylated at Y783. For negative controls, treat samples with lambda phosphatase or use non-phosphorylated peptide competition to confirm specificity.
- In Western blot, the predicted molecular weight of PLCG1 is ~149 kDa. Ensure adequate separation of high-molecular-weight proteins using appropriate gel percentage (e.g., 6-8% SDS-PAGE) and transfer conditions.
- For ELISA, the antibody can be used as a detection antibody in a phospho-specific sandwich ELISA format. Validate in the relevant sample system, such as stimulated vs. unstimulated cell lysates.
- When detecting endogenous PLCG1 phosphorylation, consider stimulating cells with growth factors (e.g., EGF, PDGF) known to activate receptor tyrosine kinases that phosphorylate PLCG1 at Y783.
- Cross-reactivity with mouse is indicated; however, confirm reactivity in your specific experimental model and validate the antibody for other species if needed.
- Since the antibody is a rabbit polyclonal, batch-to-batch variability might occur; include appropriate controls and titrate for optimal concentration.
