Biological Background: PKC alpha/beta II Function and Localization

  • Protein kinase C alpha type (PKC alpha) is a calcium-activated, phospholipid- and diacylglycerol-dependent serine/threonine-protein kinase encoded by the PRKCA gene. It plays pivotal roles in regulating cell proliferation, apoptosis, differentiation, migration, adhesion, tumorigenesis, cardiac hypertrophy, angiogenesis, platelet function, and inflammation.
  • PKC alpha localizes to the cytoplasm, cell membrane, mitochondrion membrane, and nucleus, dynamically translocating to specific compartments upon activation to exert its diverse functions.
  • During chemokine-induced CD4+ T cell migration, PKC alpha phosphorylates the CDC42-guanine exchange factor DOCK8, leading to its dissociation from LRCH1 and subsequent activation of GTPase CDC42, thereby promoting cell motility. Related references: PMID:28028151

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic phosphorylated peptide around T638 of human PKCalpha/beta II. Consider validating the antibody in your experimental system by treating cells with PMA or other PKC activators to enhance phosphorylation at T638/641 prior to analysis.
  • For Western blotting, use appropriate phosphatase inhibitors during sample preparation to preserve the phospho-epitope. A band around 76–77 kDa is expected.
  • In immunohistochemistry (IHC-P), perform antigen retrieval as required and include phosphatase inhibitors in incubation steps to maintain phosphorylation signals.
  • For immunofluorescence/immunocytochemistry (IF/ICC), monitor translocation of phosphorylated PKC from cytoplasm to membrane upon activation as a positive control.
  • In ELISA, validate the specificity using both phosphorylated and non-phosphorylated peptides to confirm phospho-specificity.
