Biological Background: Serine/threonine-protein Kinase D1 (PRKD1) Function and Localization
- Serine/threonine-protein kinase D1 (PRKD1), also known as PKC mu or nPKC-D1, is a member of the protein kinase D family that responds to diacylglycerol signals.
- Converts transient diacylglycerol (DAG) signals into sustained physiological responses by regulating MAPK8/JNK1, Ras signaling, Golgi membrane integrity, and NF-kappa-B-mediated cell survival. Related references: PMID:10764790 PMID:12505989 PMID:12637538
- Is critical for cell migration, osteoblast differentiation via HDAC7 nuclear export, and neuronal polarity maintenance. Related references: PMID:18509061
- Mediates cardiac hypertrophy through HDAC5 phosphorylation and pathological remodeling, and promotes VEGFA-induced angiogenesis via HDAC7 phosphorylation. Related references: PMID:18332134 PMID:19211839
- Under genotoxic stress, is cleaved by caspase-3 to activate kinase function and enhance apoptosis; also involved in flagellin-stimulated inflammatory responses by phosphorylating TLR5. Related references: PMID:10764790 PMID:17442957
- Phosphorylates EGFR on threonine residues, leading to suppression of EGF-induced MAPK8/JNK1 activation. Related references: PMID:10523301
- Localizes to cytoplasm, cell membrane, and trans-Golgi network; under oxidative stress, is phosphorylated at Tyr-463 via SRC-ABL1 to promote cell survival. Related references: PMID:12505989
Experimental Guidance and Technical Tips
- Use phosphatase inhibitors during sample preparation to preserve the phosphorylation at Ser916.
- Validate the antibody in your cell line or tissue of interest; consider using a pan-PRKD1 antibody for normalization.
- For Western blot, a band around 102 kDa is expected; optimize blocking and antibody dilutions.
- For ELISA, the antibody may be used as a capture or detection reagent; titrate for optimal performance.
- Note that this antibody was raised against a peptide corresponding to human PRKD1 Ser912 (homologous to Ser916), so cross-reactivity with other phospho-sites or species should be verified.
