Biological Background: PKC delta Function and Localization

  • PKC delta, encoded by PRKCD (also known as nPKC-delta, MAY1, ALPS3, CVID9), is a calcium-independent, phospholipid- and diacylglycerol-dependent serine/threonine-protein kinase.
  • Plays dual roles in cell death, acting as a pro-apoptotic kinase in DNA damage-induced apoptosis and an anti-apoptotic kinase in cytokine receptor-initiated cell death; also functions as a tumor suppressor and promotes survival in certain cancers. Related references: PMID:21406692 PMID:21810427 PMID:15774464
  • Upon DNA damage, mediates p53/TP53 transcription activation, BAX/BAK mitochondrial apoptosis, and MCL1 degradation; it also regulates cell cycle progression by upregulating CDKN1A/p21 and inhibiting cyclin A2, causing G1/S arrest, and phosphorylates CDK1 for G2/M checkpoint. Related references: PMID:21406692 PMID:21810427
  • Regulates NADPH oxidase activity and reactive oxygen species production in neutrophils and eosinophils by direct phosphorylation of NCF1/p47-phox and through MAPK1/3 (ERK1/2) signaling. Related references: PMID:19801500 PMID:11748588
  • In platelets, acts as a negative regulator of filopodia formation and actin polymerization by interacting with and negatively regulating VASP phosphorylation; also differentially regulates dense granule secretion downstream of PAR and CD36 receptors. Related references: PMID:16940418 PMID:19587372
  • Subcellular localization includes cytoplasm, perinuclear region, nucleus, cell membrane, mitochondria, and endomembrane system, reflecting its diverse signaling roles.
  • PKC delta is a phosphoprotein; phosphorylation at tyrosine residues (including Tyr311) modulates its kinase activity, protein interactions, and subcellular redistribution.
  • Phosphorylates multiple substrates such as MUC1, 14-3-3 proteins, ELAVL1, PLSCR3, SMPD1, and GLI1, regulating beta-catenin signaling, mRNA stability, cardiolipin exposure, sphingomyelin secretion, and smoothened signaling. Related references: PMID:11877440 PMID:18285462 PMID:12649167 PMID:17303575 PMID:19015273

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic phosphorylated peptide corresponding to Tyr311 of rat PKC delta; this sequence is conserved in human and mouse PKC delta, supporting cross-reactivity.
  • For Western blot analysis, consider testing a range of antibody dilutions and using phosphatase inhibitors during cell lysis to preserve phosphorylation. Validate in the relevant sample system, such as cell lysates treated with inducers of DNA damage or PMA to monitor PKC delta Tyr311 phosphorylation.
  • For ELISA applications, consider using the antibody for detection of phosphorylated PKC delta in cell or tissue extracts. Include appropriate controls with non-phosphorylated peptide to confirm phosphorylation-specific binding.
  • Since the antibody reacts with human and mouse, it can be used for cross-species studies; verify performance in your specific experimental context.
