Biological Background: PKC alpha and PKC delta Function and Localization
- PRKCA (PKC alpha) is a calcium-activated, phospholipid- and DAG-dependent serine/threonine-protein kinase of the PKC family, which also includes PRKCD (PKC delta). It regulates cell proliferation, apoptosis, differentiation, migration, and adhesion, and is highly expressed in various cancers, acting as a tumor promoter.
- PRKCA phosphorylates key substrates such as RAF1, BCL2, and TNNT2, thereby modulating the MAPK/ERK signaling cascade and apoptosis. For instance, phosphorylation of BCL2 promotes cell survival in leukemia cells.
- PRKCA plays a critical role in cell motility: upon wounding, it translocates to lamellipodia and phosphorylates CSPG4, inducing cytoskeletal rearrangement and polarized migration.
