Biological Background: PKC alpha Function and Localization

  • Protein kinase C alpha type (PKC alpha) is a calcium-activated, phospholipid- and diacylglycerol (DAG)-dependent serine/threonine-protein kinase that regulates cell proliferation, apoptosis, differentiation, migration, adhesion, tumorigenesis, cardiac hypertrophy, angiogenesis, platelet function, and inflammation.
  • Plays dual roles in cell growth: promotes proliferation via RAF1/MAPK/ERK signaling and can induce cell cycle arrest through hyperphosphorylation of RB1 and upregulation of CDK inhibitors CDKN1A and CDKN1B in certain contexts.
  • Exhibits anti-apoptotic functions in cancer cells by suppressing p53/TP53-mediated IGFBP3 activation and phosphorylating BCL2 in leukemia, contributing to tumor promoter activity in gliomas and breast cancers.
  • In CD4+ T cell chemokine-induced migration, phosphorylates DOCK8, causing its dissociation from LRCH1 and activation of CDC42 GTPase. Related references: PMID:28028151
  • In the kidney, phosphorylates KLHL3 in response to angiotensin II signaling, disrupting the KLHL3-WNK4 interaction and modulating ion homeostasis. Related references: PMID:25313067
  • Phosphorylates and activates LRRK1, which in turn phosphorylates RAB proteins involved in intracellular trafficking. Related references: PMID:36040231
  • Subcellular localization includes cytoplasm, cell membrane, mitochondrion membrane, and nucleus, enabling signal transduction at multiple sites.
  • Classified as a proto-oncogene and phosphoprotein with zinc-finger domains, belonging to the AGC kinase family.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic phosphorylated peptide encompassing residues around S657 and Y658; consider verifying phospho-specificity by treating samples with phosphatase or using non-phosphorylated peptide competition.
  • For Western blotting, the predicted molecular weight is 77 kDa; validate band specificity in the relevant cell or tissue lysate system, possibly after stimulation with known PKC activators (e.g., PMA).
  • In ELISA applications, optimize coating antigen and antibody dilutions; cross-reactivity is reported for human, but testing on other species may be needed if conservation of the phosphorylation site is high.
