Biological Background: Astrocytic Phosphoprotein PEA-15 Function and Localization

  • PEA-15 (Phosphoprotein Enriched in Astrocytes) is a 15 kDa cytoplasmic protein encoded by the PEA15 gene. Alternative splicing produces multiple isoforms.
  • It acts as a multifunctional adaptor, blocking Ras-mediated inhibition of integrin activation and modulating the ERK MAP kinase cascade.
  • PEA-15 inhibits apoptosis by suppressing both TNFRSF6- and TNFRSF1A-mediated CASP8 activation.
  • It retains RPS6KA3 in the cytoplasm, thereby inhibiting its activities (By similarity).
  • The protein plays a key role in glucose transport by regulating the plasma membrane levels of SLC2A1 and the insulin-dependent trafficking of SLC2A4.
  • Ubiquitously expressed, with highest levels in heart, brain, muscle, and adipose tissue; expression is elevated in tissues from individuals with type 2 diabetes.
  • The S104 phosphorylation site is critical for modulating PEA-15 function.
  • Keywords: Apoptosis, Sugar transport, Phosphoprotein, Alternative splicing, Cytoplasm.

Experimental Guidance and Technical Tips

  • This antibody was generated against a synthetic phosphopeptide corresponding to residues surrounding S104 of human PEA-15; it is expected to specifically recognize the phosphorylated form.
  • For Western blot, a band around 15 kDa is anticipated; confirm band specificity by including phosphatase-treated controls.
  • When performing ELISA, coat plates with the phosphopeptide antigen or cell lysates; validate binding specificity under non-denaturing conditions.
  • Cross-reactivity with mouse and rat is predicted based on sequence conservation; verify in the relevant sample system before extended studies.
  • Include phosphatase inhibitors during sample preparation to preserve phosphorylation signals.
