Biological Background: PEA15 Function and Localization
- Astrocytic phosphoprotein PEA-15 (also known as 15 kDa phosphoprotein enriched in astrocytes or Phosphoprotein enriched in diabetes, PED) is a 15 kDa cytoplasmic protein encoded by the PEA15 gene.
- PEA-15 blocks Ras-mediated inhibition of integrin activation and modulates the ERK MAP kinase cascade. It inhibits RPS6KA3 activities by retaining it in the cytoplasm.
- It inhibits both TNFRSF6- and TNFRSF1A-mediated CASP8 activity and apoptosis, playing a role in cell survival.
- PEA-15 regulates glucose transport by controlling the plasma membrane content of SLC2A1 glucose transporters and the insulin-dependent trafficking of SLC2A4.
- Ubiquitously expressed, with highest levels in heart, brain, muscle, and adipose tissue; lower in glucose-producing tissues; expression is elevated in type 2 diabetes.
- Classified as a phosphoprotein, with a phosphorylation site at Ser104 (as targeted by this antibody), and involved in apoptosis and sugar transport.
- Alternative splicing produces isoforms, and the protein has been identified by direct protein sequencing and proteomics.
Experimental Guidance and Technical Tips
- For Western Blot (WB), due to the low molecular weight (15 kDa), consider using a 12-15% SDS-PAGE gel and optimize transfer conditions for small proteins.
- Since the antibody targets a phosphorylation site (S104), validate its phospho-specificity by treating cell lysates with phosphatase or using non-phosphorylated peptide competition assays.
- In IHC-P applications, standard antigen retrieval methods (e.g., citrate buffer, pH 6.0) may be applied; inclusion of both positive and negative controls is recommended.
- The listed cross-reactivity with human, mouse, and rat indicates that the antibody may be suitable for multiple species, but independent validation in each model is advised.
- As a polyclonal antibody, lot-to-lot consistency may vary; consider pre-testing a small aliquot before large-scale experiments.
