Biological Background: Platelet-derived growth factor receptor alpha Function and Localization

• Platelet-derived growth factor receptor alpha (PDGFRA, also known as CD140a or PDGFR-2) is a cell-surface tyrosine kinase receptor belonging to the PDGFR family. It binds PDGFA, PDGFB, and PDGFC ligands, mediating essential signals for cell proliferation, survival, migration, and differentiation.
• PDGFRA plays a critical role in embryonic development, including normal skeleton formation, cephalic closure, and development of the gastrointestinal mucosa and intestinal villi. It is also required for the recruitment of mesenchymal cells.
• In wound healing and platelet activation, PDGFRA signaling promotes cell migration and chemotaxis, and contributes to thrombin-induced platelet aggregation and secretion of granule agonists.
• The receptor localizes to the cell membrane, as well as to cell projections such as cilia, and the Golgi apparatus. Its subcellular distribution is regulated by ligand binding and activation.
• PDGFRA undergoes autophosphorylation at multiple tyrosine residues, including Y1018, which creates docking sites for downstream signaling molecules like PLCG1, PIK3R1, and PTPN11, leading to activation of MAPK/ERK, PI3K/AKT, and STAT pathways.
• Tissue-specific expression is widespread; detected in brain, fibroblasts, smooth muscle, heart, and embryos, as well as in primary and metastatic colon tumors. It is also found in platelets at the protein level.
• Post-translational modifications include phosphorylation, glycosylation, and ubiquitination, which regulate receptor activity, trafficking, and degradation.

• The immunogen corresponds to a phosphorylated peptide around tyrosine 1018 (Y1018) in the intracellular domain of human PDGFRA. This site is a known autophosphorylation site critical for downstream signaling.
• The antibody is suitable for Western Blot (WB) and ELISA applications, as indicated. For WB, consider using appropriate positive controls (e.g., stimulated cell lysates where PDGFRA is phosphorylated) and validate the specificity of phosphorylation detection by comparing treated vs untreated samples.
• Since PDGFRA is a transmembrane receptor, membrane protein extraction protocols may enhance detection. The predicted molecular weight is 123 kDa; consider using appropriate gel percentages and transfer conditions for high-molecular-weight proteins.
