Biological Background: Platelet-derived growth factor receptor alpha Function and Localization

  • Platelet-derived growth factor receptor alpha (PDGFRA, also known as CD140a or PDGFR-2) is a cell-surface tyrosine kinase receptor belonging to the PDGFR family. It binds PDGFA, PDGFB, and PDGFC ligands, mediating essential signals for cell proliferation, survival, migration, and differentiation.
  • PDGFRA plays a critical role in embryonic development, including normal skeleton formation, cephalic closure, and development of the gastrointestinal mucosa and intestinal villi. It is also required for the recruitment of mesenchymal cells.
  • In wound healing and platelet activation, PDGFRA signaling promotes cell migration and chemotaxis, and contributes to thrombin-induced platelet aggregation and secretion of granule agonists.

  • The immunogen corresponds to a phosphorylated peptide around tyrosine 1018 (Y1018) in the intracellular domain of human PDGFRA. This site is a known autophosphorylation site critical for downstream signaling.
  • The antibody is suitable for Western Blot (WB) and ELISA applications, as indicated. For WB, consider using appropriate positive controls (e.g., stimulated cell lysates where PDGFRA is phosphorylated) and validate the specificity of phosphorylation detection by comparing treated vs untreated samples.
