Biological Background: p90Rsk (RPS6KA1) Function and Localization
- Protein identity: Ribosomal protein S6 kinase alpha-1, also known as p90-RSK 1, RSK-1, MAPKAPK1A, is encoded by the RPS6KA1 gene. It belongs to the RSK family of serine/threonine-protein kinases.
- Core signaling function: Acts downstream of ERK/MAPK signaling to mediate mitogenic and stress responses. It activates transcription factors CREB1, ETV1, and NR4A1, and regulates cell proliferation, survival, and differentiation via mTOR and apoptotic pathways. Experimental Guidance and Technical Tips
- This antibody was generated using a synthetic phosphorylated peptide corresponding to residues around T359 and S363 of human RSK1. It is expected to specifically recognize the doubly phosphorylated form.
- For Western blot (WB), a band at approximately 83 kDa is anticipated. Use appropriate molecular weight markers.
- To preserve phosphorylation, include phosphatase inhibitors in lysis buffers. For negative control, consider phosphatase treatment of lysates.
- For immunoprecipitation (IP) and immunofluorescence (IF/ICC), optimize antibody concentration and fixation conditions for each experimental system.
- The antibody reacts with human, mouse, and rat targets, but sequence alignment of the phosphopeptide across these species is recommended for cross-reactivity confidence.
