- RPS6KB1 encodes the 70 kDa ribosomal protein S6 kinase beta-1 (p70S6K), also known as S6K1, p70 S6 kinase alpha, or serine/threonine-protein kinase 14A (STK14A).
- The kinase acts as a key downstream effector of mTORC1, regulating protein synthesis and cell growth. Upon mitogenic stimulation, mTORC1-mediated phosphorylation dissociates p70S6K from the EIF3 complex, enabling it to phosphorylate translation factors such as EIF4B and PDCD4, thereby promoting cap-dependent translation initiation. - p70S6K1 contributes to cell survival by inactivating the pro-apoptotic protein BAD through phosphorylation. Additionally, it phosphorylates mitochondrial URI1, disrupting the URI1-PPP1CC complex; released PPP1CC dephosphorylates p70S6K1 at Thr-412, creating a negative feedback loop for anti-apoptotic signaling.
