Biological Background: p38 MAPK Function and Localization
- MAPK11 is a serine/threonine kinase and a member of the p38 mitogen-activated protein kinase family, also known as SAPK2b or p38-2.
- As part of the MAP kinase signal transduction pathway, it responds to extracellular stimuli such as pro-inflammatory cytokines and physical stress, leading to direct activation of transcription factors. Related references: PMID:12452429, PMID:20626350, PMID:35857590
- p38 MAPKs phosphorylate a broad range of downstream targets, including kinases MSK1/2, MK2/3, and MNK1/2, which in turn regulate transcription, mRNA stability, and translation. Related references: PMID:9687510, PMID:11154262
- In the nucleus, p38 MAPKs phosphorylate transcription factors like ATF1, ATF2, ATF6, ELK1, PTPRH, DDIT3, p53, MEF2C, and MEF2A to modulate gene expression. Related references: PMID:10330143, PMID:15356147, PMID:9430721
- The kinase also regulates chromatin remodeling by inducing phosphorylation of histone H3 at 'Ser-10', enhancing NF-kappa-B recruitment to promoters of inflammatory genes.
- Subcellularly, MAPK11 localizes to both the cytoplasm and nucleus.
- Tissue expression is highest in the brain and heart, with additional presence in placenta, lung, liver, skeletal muscle, kidney, and pancreas.
- Key post-translational modifications include phosphorylation at T180 and Y182 (critical for activation), and the protein is classified under stress response and transcription regulation keywords.
Experimental Guidance and Technical Tips
- Since the immunogen is a synthetic phosphorylated peptide, the antibody specifically recognizes the activated, dual-phosphorylated form of p38 MAPK. Verify specificity by comparing treated versus untreated cell lysates (e.g., with and without stress inducers).
- For Western blotting, consider using a 10% SDS-PAGE gel and loading 20–30 μg of protein lysate. Multiple bands may be observed due to detection of different p38 isoforms (predicted weights: 29, 34, 35, and 41 kDa). Include appropriate molecular weight markers and a loading control.
- In immunoprecipitation or ELISA applications, confirm phospho-specific binding under non-denaturing conditions to ensure epitope accessibility.
- For immunofluorescence (IF-P), use a fixation method that preserves phosphorylation status (e.g., 4% paraformaldehyde) and permeabilize with 0.1% Triton X-100.
- For immunohistochemistry (IHC-P), antigen retrieval with citrate buffer (pH 6.0) may be required. Titrate antibody to find optimal dilution.
- Always include a negative control by pre-adsorbing the antibody with the phosphorylated peptide, or by using alkaline phosphatase treatment to confirm phospho-specificity.
