Biological Background: NFKB1 Function and Localization
- NFKB1 encodes the p105 precursor which is processed into the active p50 subunit of NF-kappa-B. P105 acts as a cytoplasmic retention factor for attached NF-kappa-B proteins. Related references: PMID:1423592
- The active p50 subunit associates with RELA/p65 to form the NF-kappa-B p65-p50 complex, a transcription factor that binds kappa-B sites in DNA. Related references: PMID:1740106 PMID:7830764
- NF-kappa-B is a pleiotropic transcription factor involved in inflammation, immunity, cell growth, and apoptosis, existing as various dimers with distinct transcriptional activities.
- The p50-p50 homodimer acts as a transcriptional repressor, but can become an activator when associated with BCL3.
- Subcellular location is primarily cytoplasmic; upon activation, NF-kappa-B complexes translocate to the nucleus.
- Key post-translational modifications include acetylation, phosphorylation, and ubiquitination, regulating its activity.
- Serine 927 phosphorylation may modulate NFKB1 function; the antibody is specific for this modification.
- Gene family: NFKB1 is a member of the NF-kappa-B family, which includes RELA, RELB, REL, and NFKB2.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic phosphopeptide corresponding to serine 927 of human NFKB1. For Western blot, consider using appropriate phosphatase inhibitors during sample preparation to preserve the phosphorylation.
- The antibody is generated in rabbit and is expected to be specific for the phosphorylated form; it may also recognize the non-phosphorylated form if present abundantly. Validate in relevant positive and negative control lysates.
- For ELISA, the peptide can be used as a coating antigen or competitor to confirm specificity.
- Since the protein weight is 105 kDa, ensure adequate gel percentage for separation (e.g., 8–10% SDS-PAGE) and use appropriate molecular weight markers.
- Cross-reactivity is reported for human; if using in other species, sequence alignment around S927 is recommended.
