- NF-κB p65 (also known as RelA, NFKB3) is a subunit of the NF-κB transcription factor complex, belonging to the Rel homology domain-containing protein family that includes RELA, RELB, REL, NFKB1/p105, and NFKB2/p52.
- The canonical NF-κB heterodimer RELA-NFKB1 binds κB DNA sites to activate genes involved in inflammation, immunity, cell growth, differentiation, and apoptosis.
- p65/RelA forms both activating heterodimers (e.g., RELA-NFKB1) and homodimers (RELA-RELA), the latter involved in invasin-mediated IL-8 expression.
- Subcellular localization is primarily cytoplasmic in an inactive state bound to IκB inhibitors; upon activation, it translocates to the nucleus to regulate transcription.
- p65 is subject to extensive post-translational modifications including phosphorylation, acetylation, methylation, ubiquitination, and S-nitrosylation, which modulate its activity and stability.
