Biological Background: NF-κB p105 Function and Localization
- NFKB1 encodes the p105 precursor protein, which is proteolytically processed to generate the active p50 subunit of the NF-κB transcription factor complex (PubMed:1423592). Related references: PMID:1423592
- The p50 subunit associates with RELA/p65 to form the p65-p50 heterodimer, the most abundant NF-κB complex, which binds κB sites (5'-GGRNNYYCC-3') in target gene promoters to activate transcription (PubMed:1740106, PubMed:7830764). Related references: PMID:1740106, PMID:7830764
- NF-κB is a pleiotropic transcription factor central to immune response, inflammation, apoptosis, and cell proliferation, regulated by IκB inhibitor proteins and diverse stimuli.
- The p105 protein contains an N-terminal Rel homology domain (RHD) and C-terminal ankyrin repeats; it functions as a cytoplasmic retention factor for NF-κB dimers, sequestering them in an inactive state.
- Upon activation signals, IκB kinases (IKKs) phosphorylate IκB, leading to its degradation and release of NF-κB dimers, which then translocate to the nucleus to modulate gene expression.
- p50 homodimers act as transcriptional repressors unless they associate with BCL3, highlighting the dimer-specific regulatory versatility of NF-κB.
- Post-translational modifications including phosphorylation, acetylation, ubiquitination, and S-nitrosylation finely tune NF-κB activity; phosphorylation at serine residues (e.g., S932) is implicated in p105 processing and activation.
- Subcellular localization is predominantly cytoplasmic for p105, while active p50-containing dimers shuttle between cytoplasm and nucleus.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic phosphopeptide corresponding to residues around Ser932 of human NF-κB p105. For Western blot, expect a major band at approximately 105 kDa (p105) and possibly a band at around 50 kDa (p50). Validate phosphorylation specificity by treating lysates with phosphatase or comparing with a non-phospho-specific antibody.
- For ELISA applications, titer and optimize the antibody concentration using phosphorylated peptide or stimulated cell lysates. Include appropriate blocking steps to reduce non-specific binding.
- Given the antibody's cross-reactivity with mouse and rat, consider using relevant cell lines (e.g., Jurkat, HeLa, RAW 264.7) as positive controls. Induction of NF-κB signaling with TNF-α, LPS, or PMA/ionomycin can enhance detection.
- Always include molecular weight markers and appropriate controls to confirm specificity, as NF-κB family members share similarity.
