Biological Background: Myosin Light Chain 2 Function and Localization
- Encodes myosin regulatory light polypeptide 9 (MYL9), also known as LC20, MLC-2C, or MRLC1; alternative splicing generates multiple isoforms.
- Plays a key role in regulation of both smooth muscle and nonmuscle cell contractile activity via its phosphorylation, and is implicated in cytokinesis, receptor capping, and cell locomotion (PubMed:11942626, PubMed:2526655). Related references: PMID:11942626 PMID:2526655
- In myoblasts, may regulate PIEZO1-dependent cortical actomyosin assembly involved in myotube formation (By similarity).
- Localizes to the cytoplasm, cytoskeleton, and cell cortex.
- Expressed in smooth muscle tissues and in some, but not all, nonmuscle cells.
- Subject to critical post-translational modifications including phosphorylation (e.g., at T18/S19), acetylation, and calcium/metal binding.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic phosphopeptide spanning T18/S19; verify phosphorylation specificity by comparing signals in native versus phosphatase-treated lysates.
- Suitable for Western blot (predicted band at ~20 kDa) and ELISA; consider using known phospho-myosin regulators as controls.
- Validate antibody performance in human smooth muscle or nonmuscle cell models relevant to your study.
