Biological Background: MYL9 / Myosin Light Chain 2 Function and Localization
- MYL9 encodes myosin regulatory light polypeptide 9, also known as 20 kDa myosin light chain (LC20), MLC-2C, myosin regulatory light chain 2 smooth muscle isoform, myosin regulatory light chain MRLC1.
- Functions as a myosin regulatory subunit crucial for regulating both smooth muscle and nonmuscle cell contractile activity via its phosphorylation. Implicated in cytokinesis, receptor capping, and cell locomotion (PubMed:11942626, PubMed:2526655). Related references: PMID:11942626 PMID:2526655
- May regulate PIEZO1-dependent cortical actomyosin assembly involved in myotube formation in myoblasts (By similarity).
- Localizes to the cytoplasm, cytoskeleton, and cell cortex.
- Expressed in smooth muscle tissues and also in some, but not all, nonmuscle cells.
- Contains calcium-binding EF-hand motifs and is subject to regulatory phosphorylation; classified as a metal-binding, actin-associated motor protein.
- Alternative splicing yields multiple isoforms.

- The immunogen is a synthetic phosphopeptide corresponding to phosphorylated T18 of human MYL9. To preserve the phosphorylation signal, we recommend using phosphatase inhibitors in lysis buffers and performing rapid sample processing.
- For Western blotting, the predicted molecular weight is approximately 20 kDa; optimize blocking and antibody incubation conditions for phospho-specific detection.
- In ELISA, consider using a non-phosphorylated control peptide to confirm phosphorylation-specific binding.
