Biological Background: MEK1/MEK2 Function and Localization
- Dual specificity protein kinases MEK1 (MAP2K1) and MEK2 (MAP2K2) are essential components of the MAPK/ERK signaling cascade, transmitting signals from activated RAS-RAF to ERK1/2.
- Upon activation, MEK1/2 catalyze the concomitant phosphorylation of a threonine and tyrosine residue in the Thr-Glu-Tyr motif of ERK1 (MAPK3) and ERK2 (MAPK1), leading to their activation.
- MEK1/2 are activated by RAF kinases (e.g., RAF1) downstream of growth factor, cytokine, and hormone receptor stimulation, linking extracellular signals to intracellular responses.

- For western blotting, MEK1/2 appear as bands around 40–44 kDa. Use appropriate positive controls (e.g., EGF-stimulated cell lysates) and include phosphatase inhibitors during sample preparation.
- For immunoprecipitation, a rabbit monoclonal antibody offers high specificity and compatibility with standard IP protocols. Validate pull-down efficiency with phospho-ERK1/2 readouts.
