Biological Background: MEK1 Function and Localization

  • Dual specificity protein kinase that is an essential component of the MAP kinase signal transduction pathway, catalyzing the concomitant phosphorylation of a threonine and a tyrosine residue in ERK1/2 (MAPK3/1) on a Thr-Glu-Tyr sequence, leading to their activation.
  • Mediates diverse biological functions including cell growth, adhesion, survival and differentiation, predominantly through regulation of transcription, metabolism and cytoskeletal rearrangements.
  • In the MAPK/ERK cascade, RAF1 activates MAP2K1/MEK1, which then phosphorylates and activates ERK1/2.
  • Activates BRAF in a KSR1 or KSR2-dependent manner, releasing inhibitory intramolecular interactions in KSR proteins and promoting KSR-BRAF dimerization and BRAF activation. Related references: PMID:29433126

  • Validated applications include Western blot (WB) and ELISA; for WB, a band at approximately 43 kDa is expected. Cross-reactivity has been observed with human, mouse, and rat MEK1.
  • Consider validating the antibody in your cell line or tissue of interest under stimulated (e.g., growth factor-treated) versus unstimulated conditions to confirm phospho-specificity.
