Biological Background: MEK1 Function and Localization

  • MEK1 (MAP2K1) is a dual-specificity protein kinase that is a central component of the MAPK/ERK signaling pathway. It is activated downstream of RAS and RAF1 upon extracellular ligand stimulation.
  • It specifically phosphorylates both Thr and Tyr residues in the Thr-Glu-Tyr motif of ERK1 (MAPK3) and ERK2 (MAPK1), leading to their activation and signal transduction.
  • MEK1 also activates BRAF in a KSR1/2-dependent manner, promoting BRAF dimerization and enhancing MAPK cascade signaling (PubMed:29433126). Related references: PMID:29433126
  • The MAPK/ERK pathway mediates diverse biological processes, including cell growth, adhesion, survival, differentiation, transcription regulation, metabolism, and cytoskeletal rearrangement.
  • MEK1 is involved in regulating endosomal dynamics, lysosome processing, and Golgi apparatus fragmentation during mitosis.
  • The protein localizes to the cytoplasm, centrosome, spindle pole body, nucleus, and membrane.
  • MEK1 is widely expressed in tissues, with extremely low levels in the brain.
  • Keywords: Acetylation, alternative splicing, ATP-binding, phosphoprotein with serine/threonine and tyrosine kinase activity.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic phosphorylated peptide around S217 of human MEK1. For optimal detection of phosphorylated MEK1, inclusion of phosphatase inhibitors during sample preparation is strongly recommended.
  • The antibody is suitable for WB and ELISA; consider validating the optimal dilution and conditions for your specific assay system.
  • Given cross-reactivity with mouse and rat, this antibody can be used across these commonly studied species; confirm expected band at ~43 kDa (MEK1 molecular weight) in your samples.
