Biological Background: MEK1 Function and Localization

  • MEK1, encoded by the MAP2K1 gene, is a dual specificity mitogen-activated protein kinase kinase that phosphorylates both threonine and tyrosine residues on its targets ERK1 and ERK2.
  • It is a core component of the MAPK/ERK signaling cascade, transmitting signals from activated RAS and RAF to ERK, thereby regulating processes such as cell growth, adhesion, survival, and differentiation.
  • MEK1 specifically catalyzes the concomitant phosphorylation of a Thr-Glu-Tyr motif in ERK1/ERK2, leading to their activation and downstream signaling.
  • MEK1 also enhances BRAF activation in a KSR1- or KSR2-dependent manner by releasing the autoinhibitory interaction of KSR proteins and promoting KSR-BRAF dimerization. Related references: PMID:29433126
  • Subcellularly, MEK1 localizes predominantly to the cytoplasm, centrosome, spindle pole body, and can transiently enter the nucleus; it also associates with membranes.
