Biological Background: MEK1 Function and Localization
- MEK1, encoded by the MAP2K1 gene, is a dual specificity mitogen-activated protein kinase kinase that phosphorylates both threonine and tyrosine residues on its targets ERK1 and ERK2.
- It is a core component of the MAPK/ERK signaling cascade, transmitting signals from activated RAS and RAF to ERK, thereby regulating processes such as cell growth, adhesion, survival, and differentiation.
- MEK1 specifically catalyzes the concomitant phosphorylation of a Thr-Glu-Tyr motif in ERK1/ERK2, leading to their activation and downstream signaling.
- MEK1 also enhances BRAF activation in a KSR1- or KSR2-dependent manner by releasing the autoinhibitory interaction of KSR proteins and promoting KSR-BRAF dimerization. Related references: PMID:29433126
- Subcellularly, MEK1 localizes predominantly to the cytoplasm, centrosome, spindle pole body, and can transiently enter the nucleus; it also associates with membranes.
- MEK1 is widely expressed across tissues, with exceptionally low expression in the brain.
- The protein is subject to acetylation and activating phosphorylation at serine and threonine residues; it is classified as a phosphoprotein and is a reference proteome member.
- Mutations in MAP2K1 are linked to human diseases including cardiomyopathy, ectodermal dysplasia, and intellectual disability.

- This antibody was raised against a synthetic phosphorylated peptide corresponding to Ser221 of human MEK1, and is intended to detect MEK1 phosphorylated at this site.
- Validate performance in Western blotting (WB) and ELISA with appropriate phosphorylated and non-phosphorylated controls to confirm phospho-specificity.
- For WB, include phosphatase inhibitors in lysis and running buffers to preserve phosphorylation signals, and consider using total MEK1 antibody as loading control.
