Biological Background: MEF2C Function and Localization

  • MEF2C is a transcription activator that binds to the MEF2 element in regulatory regions of many muscle-specific genes, controlling cardiac morphogenesis and myogenesis.
  • It is also involved in vascular development and enhances transcriptional activation mediated by SOX18.
  • MEF2C plays an essential role in hippocampal-dependent learning and memory by regulating excitatory synapse number and synaptic transmission.
  • Crucial for normal neuronal development, distribution, and electrical activity in the neocortex, and may be involved in neurogenesis and cortical architecture (By similarity).
  • Necessary for proper development of megakaryocytes and platelets, as well as bone marrow B-lymphopoiesis.
  • Required for B-cell survival and proliferation in response to BCR stimulation, efficient IgG1 antibody responses to T-cell-dependent antigens, and normal induction of germinal center B-cells.
  • Isoforms lacking the repressor domain are more active than isoform 1 (alternative splicing).
  • Subcellular localization includes nucleus, cytoplasm, and sarcoplasm; tissue expression in brain and skeletal muscle.

Experimental Guidance and Technical Tips

  • For Western blot, treat cell lysates with lambda phosphatase to confirm phosphorylation-specific signal; expect a band around 51 kDa.
  • Validate antibody specificity in relevant human cell lines or tissues expressing MEF2C, such as neuronal or skeletal muscle cells.
  • For ELISA applications, consider using phospho-peptide and non-phospho-peptide as coating antigens to confirm phospho-specificity.
  • Since MEF2C has multiple isoforms, verify detection of the targeted isoform using appropriate controls or recombinant proteins.
  • Optimize antibody concentration for each application; recommended starting dilutions can be determined empirically.
