- Protein max (Myc-associated factor X) is a class D basic helix-loop-helix protein (bHLHd4) that functions as a transcription regulator.
- Forms sequence-specific DNA-binding heterodimers with MYC or MAD, recognizing the E-box motif 5'-CAC[GA]TG-3'. The MYC:MAX complex activates transcription, while MAD:MAX represses it.
- Can repress transcription by recruiting a chromatin remodeling complex with H3 'Lys-9' histone methyltransferase activity.
- Represses MYC transcriptional activity from E-box elements.
- Localizes to the nucleus and is also found in cell projections, specifically dendrites.
- Tissue expression is highest in brain, heart, and lung, with lower levels in liver, kidney, and skeletal muscle.
- Key post-translational modifications include phosphorylation (at Ser-11 among other sites), acetylation, and alternative splicing. Classified as a phosphoprotein and DNA-binding factor.
