- Dual specificity protein kinase acting as an essential component of the MAP kinase signal transduction pathway.
- Essential component of the stress-activated protein kinase/c-Jun N-terminal kinase (SAP/JNK) signaling pathway; directly phosphorylates and activates MAPK8/JNK1, MAPK9/JNK2, and MAPK10/JNK3.
- Along with MAP2K7/MKK7, is one of the only known kinases to directly activate JNKs; MAP2K4 preferentially phosphorylates the Tyr residue within the Thr-Pro-Tyr motif.
- Additionally activates p38 MAPKs (MAPK11, MAPK12, MAPK13, MAPK14), unlike MAP2K7 which exclusively targets JNKs.
- Required for maintaining peripheral lymphoid homeostasis.
- Participates in mitochondrial death signaling, promoting cytochrome c release and apoptosis.
- Subcellular localization includes both cytoplasm and nucleus.
- Abundantly expressed in skeletal muscle, with wide distribution across other tissues.
