Biological Background: MAP2K4 Function and Localization

  • Dual specificity protein kinase acting as an essential component of the MAP kinase signal transduction pathway.
  • Essential component of the stress-activated protein kinase/c-Jun N-terminal kinase (SAP/JNK) signaling pathway; directly phosphorylates and activates MAPK8/JNK1, MAPK9/JNK2, and MAPK10/JNK3.
  • Along with MAP2K7/MKK7, is one of the only known kinases to directly activate JNKs; MAP2K4 preferentially phosphorylates the Tyr residue within the Thr-Pro-Tyr motif.
  • Additionally activates p38 MAPKs (MAPK11, MAPK12, MAPK13, MAPK14), unlike MAP2K7 which exclusively targets JNKs.
  • Required for maintaining peripheral lymphoid homeostasis.
  • Participates in mitochondrial death signaling, promoting cytochrome c release and apoptosis.
  • Subcellular localization includes both cytoplasm and nucleus.
  • Abundantly expressed in skeletal muscle, with wide distribution across other tissues.

Experimental Guidance and Technical Tips

  • Confirm phospho-specificity by treating samples with lambda phosphatase or by co-incubation with non-phosphorylated blocking peptide.
  • Include appropriate positive and negative control lysates (e.g., stimulated versus unstimulated cells) to validate phospho-signal in WB.
  • For mouse or rat samples, verify cross-reactivity and sequence alignment at the phosphorylation site before quantitative assays.
  • Determine optimal antibody dilution for each application experimentally; typical starting dilutions can be found in the antibody datasheet.
  • In ELISA, ensure consistent plate coating and detection reagents to minimize variability in phospho-quantification.
