Biological Background: LCP2 Function and Localization
- Adapter protein primarily involved in signaling pathways within T-cells, as well as other immune cells such as platelets, mast cells, and natural killer (NK) cells. Related references: PMID:11313406, PMID:33159816
- Plays a crucial role for transducing signal from the T-cell receptor (TCR) after antigen recognition leading to T-cell activation. Once phosphorylated by ZAP70, mediates interactions with VAV1, NCK and ITK. Related references: PMID:8673706, PMID:8702662
- Stimulates the activation of PKC-theta/PRKCQ and NF-kappa-B transcriptional activity in response to CD3 and CD28 costimulation. Related references: PMID:11313406
- Plays an essential role in AGER-induced signaling pathways including p38 MAPK and ERK1/2 activation leading to cytokine release and pro-inflammatory responses. Related references: PMID:33436632
- Localizes to the cytoplasm.
- Highly expressed in spleen, thymus and peripheral blood leukocytes. Highly expressed also in T-cell and monocytic cell lines, expressed at lower level in B-cell lines. Not detected in fibroblast or neuroblastoma cell lines.
- Contains an SH2 domain and is subject to phosphorylation; annotated as a phosphoprotein and reference proteome member.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic phosphorylated peptide around Y145; consider verifying specificity using a non-phosphorylated peptide control in parallel experiments.
- For Western blot, maintain phospho-protein integrity by including phosphatase inhibitors in lysis buffers and consider using a phospho-specific detection protocol.
- In ELISA, a sandwich approach with capture and detection antibodies may enhance signal; validate the assay with known phosphorylated and non-phosphorylated antigen standards.
- Cross-reactivity with mouse LCP2 is reported; confirm expected band size (~60 kDa) and sequence homology in the relevant sample system.
