Biological Background: LCK Function and Localization
- LCK is a non-receptor tyrosine-protein kinase essential for T-cell selection, maturation in the thymus, and mature T-cell function.
Related references: PMID:2470098 - Upon T-cell receptor (TCR) engagement with peptide-MHC, LCK is recruited to the TCR-CD3 complex and phosphorylates tyrosine residues within ITAMs of CD3 and TCR chains, triggering ZAP70 recruitment and downstream lymphokine production.
Related references: PMID:2470098 PMID:40592325 - LCK also participates in signaling via CD2 and IL2 receptors: CD2 association leads to LCK hyperphosphorylation, and IL2 binding enhances LCK activity to promote T-cell proliferation.
- It phosphorylates multiple substrates including RUNX3, PTK2B/PYK2, MAPT, RHOH, and TYROBP, and interacts with FYB2.
Related references: PMID:27335501 - Localizes to the cell membrane, cytoplasm, and cytosol.
- Expressed specifically in lymphoid cells.
- Annotated with keywords such as alternative splicing, ATP-binding, kinase, phosphoprotein, proto-oncogene, SH2 and SH3 domains, and lipidation (myristate and palmitate), reflecting its multifaceted regulation.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic phosphopeptide around Tyr394, so the antibody is expected to specifically recognize LCK phosphorylated at this activation loop site.
- For WB, use phosphatase inhibitors during sample preparation to preserve phosphorylation. A band at ~58 kDa is anticipated in lymphoid cell lysates from human and mouse.
- Validate antibody performance in appropriate T-cell models (e.g., Jurkat cells) stimulated through CD3/CD28 to induce robust Tyr394 phosphorylation.
- IF/ICC may require optimized fixation/permeabilization; consider testing in activated T-cells or thymus tissue sections.
