Biological Background: LATS1/LATS2 Function and Localization
- Serine/threonine-protein kinase LATS1 (Large tumor suppressor homolog 1) is a core component of the Hippo signaling pathway, which regulates organ size and suppresses tumors by restricting proliferation and promoting apoptosis.
- In the Hippo kinase cascade, MST1/STK4 and MST2/STK3, complexed with SAV1, phosphorylate LATS1/2 in complex with MOB1; activated LATS1/2 then phosphorylates and inactivates the oncoproteins YAP1 and WWTR1/TAZ, preventing their nuclear translocation and transcription of pro-proliferative genes.
