Biological Background: JAK2 Function and Localization
- JAK2 is a non-receptor tyrosine-protein kinase (Janus kinase 2) that phosphorylates type I and type II cytokine receptors, including receptors for growth hormone, prolactin, leptin, erythropoietin, thrombopoietin, interferons, and interleukins. Related references: PMID:15690087, PMID:15899890, PMID:7615558
- Upon ligand binding, JAK2 phosphorylates cytoplasmic tyrosine residues on the receptor, creating docking sites for STAT transcription factors, which are then phosphorylated, dimerize, and translocate to the nucleus to activate gene transcription. Related references: PMID:15690087, PMID:9618263
- JAK2 is central to erythropoiesis: stimulation with erythropoietin (EPO) triggers JAK2 autophosphorylation, association with EPOR, and subsequent phosphorylation and activation of STAT5, driving transcription of essential erythroid genes. Related references: PMID:9657743
- In the nucleus, JAK2 has a chromatin regulatory role by phosphorylating histone H3 at tyrosine 41 (H3Y41ph), a modification that promotes exclusion of heterochromatin protein 1 alpha (CBX5/HP1α) from chromatin. Related references: PMID:19783980
- JAK2 phosphorylates CDKN1B to regulate cell cycle, mediates angiotensin-2-induced ARHGEF1 phosphorylation, and cooperates with TEC for reciprocal phosphorylation to activate FOS transcription. Related references: PMID:21423214, PMID:20098430
- Subcellular localization: JAK2 is associated with the endomembrane system, cytoplasm, and nucleus.
- Tissue specificity: JAK2 is ubiquitously expressed throughout most tissues.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic phosphorylated peptide around Y221 of human JAK2; thus, the antibody is specific for the phosphorylated form. For Western blot, include phosphatase inhibitors in lysis buffer and consider phosphatase treatment as a negative control.
- For ELISA, validate the antibody's specificity using phosphorylated versus non-phosphorylated peptide competitors or relevant recombinant proteins.
- Cross-reactivity is listed for human, mouse, and rat; verify reactivity in your specific sample system and consider sequence alignment if extending to other species.
