Biological Background: IKKγ (NEMO) Function and Localization
- IKKγ (NEMO, IKBKG) is the regulatory subunit of the IKK core complex, which phosphorylates NF-kB inhibitors, leading to inhibitor degradation and NF-kB activation. Related references: PMID:14695475, PMID:20724660, PMID:21518757
- It binds scaffolding polyubiquitin chains, recognizing both 'Lys-63'-linked and linear polyubiquitin with high affinity for linear chains, playing a key role in IKK activation by multiple signaling receptors. Related references: PMID:16547522, PMID:18287044, PMID:19033441
- IKKγ is implicated in NF-kB-mediated protection from cytokine toxicity and is essential for viral activation of IRF3. Related references: PMID:19854139
- Involved in antiviral innate immune responses triggered by TLR3 and IFIH1, requiring 'Lys-27'-linked polyubiquitination. Related references: PMID:20724660
- Also functions as a mediator for HTLV-1 Tax oncoprotein activation of NF-kB.
- Subcellular localization includes both the cytoplasm and nucleus.
- Expressed in a wide range of tissues, including heart, brain, placenta, lung, liver, skeletal muscle, kidney, and pancreas.
- Contains a coiled coil domain and a zinc finger domain; mutations in the gene are associated with ectodermal dysplasia and osteopetrosis.
Experimental Guidance and Technical Tips
- This antibody is raised against a synthetic phosphopeptide corresponding to residues surrounding Ser376 of human IKKγ. For optimal phospho-specific detection, include phosphatase inhibitors in lysis buffers and consider using phosphatase treatment as a negative control.
- The antibody has been validated for Western blot (WB) and ELISA; for other applications, we recommend pilot experiments to determine appropriate dilutions and conditions.
- Predicts bands at approximately 48 kDa, 56 kDa, and 37 kDa; multiple bands may reflect alternative splicing or post-translational modifications.
- Expected to cross-react with human, mouse, and rat IKKγ based on sequence homology; for other species, verify epitope conservation.
