Biological Background: NF-kappa-B inhibitor alpha Function and Localization
- NF-kappa-B inhibitor alpha (IκBα, also known as I-kappa-B-alpha, IkB-alpha, IkappaBalpha, MAD3) is encoded by the NFKBIA gene (IKBA, MAD3, NFKBI) and acts as a critical regulator of the NF-κB transcription factor family.
- It inhibits NF-κB activity by trapping RELA/p65 and NFKB1/p50 dimers in the cytoplasm, masking their nuclear localization signals (PubMed:1493333, PubMed:36651806, PubMed:7479976). Related references: PMID:1493333, PMID:36651806, PMID:7479976
- Upon cellular stimulation by immune and pro-inflammatory signals, IκBα undergoes phosphorylation, which promotes its ubiquitination and degradation, allowing NF-κB dimers to translocate to the nucleus and activate transcription (PubMed:7479976, PubMed:7628694, PubMed:7796813). Related references: PMID:7479976, PMID:7628694, PMID:7796813
- Subcellularly, IκBα primarily localizes to the cytoplasm, but it can also be found in the nucleus under certain conditions, reflecting its dynamic role in shuttling NF-κB complexes.
- The protein is subject to various post-translational modifications, including phosphorylation, ubiquitination, hydroxylation, and isopeptide bond formation, which regulate its stability and function.
- IκBα is associated with disease variants linked to ectodermal dysplasia, highlighting its role in developmental and immune-related disorders.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic phosphopeptide corresponding to residues surrounding tyrosine 42 of human IκBα; for Western blot, consider using phosphatase inhibitors during sample preparation to preserve phosphorylation.
- Validate antibody specificity in your sample system by including controls (e.g., non-phosphorylated peptide competition or phosphatase-treated lysates).
- For WB, a band around 36 kDa is expected; optimize blocking and antibody dilution conditions for your specific experimental setup.
