Biological Background: Phospho-IGF1R/INSR Function and Localization
- Insulin-like growth factor 1 receptor (IGF1R) is a receptor tyrosine kinase that binds IGF1 with high affinity, and IGF2 and insulin with lower affinity. It plays a pivotal role in cell growth, survival, and is crucial for tumor transformation and malignant cell survival.
- Ligand binding triggers receptor autophosphorylation on tyrosine residues, including Y1165 and Y1166 in the activation loop, leading to recruitment and tyrosine phosphorylation of adaptor proteins such as IRS1/2, Shc, and 14-3-3 proteins.
- Phosphorylated IRS1 activates two major signaling cascades: the PI3K-AKT/PKB pathway, which inhibits apoptosis and promotes protein synthesis via mTOR, and the Ras-MAPK pathway, which drives cellular proliferation.
- IGF1R also signals through the JAK/STAT pathway; activation of STAT3 may be essential for the receptor’s transforming activity. Additionally, JNK kinases can be activated, and IGF1 exerts inhibitory effects on JNK via ASK1.

- Subcellularly, IGF1R is localized to the cell membrane.
- It is expressed in a wide range of tissues and often overexpressed in tumors, including melanomas and cancers of the colon, pancreas, prostate, and kidney. As a hybrid receptor with INSR, it is found in muscle, heart, kidney, adipose tissue, and other sites.
- Post-translational modifications include autophosphorylation, cleavage on basic residues, disulfide bond formation, glycosylation, and ubiquitination. The calculated molecular weight of the unprocessed precursor is approximately 154 kDa.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic phosphorylated peptide spanning the activation loop phosphorylation sites (Y1165/Y1166) of human IGF1R, which are conserved in INSR (Y1189/Y1190). This design targets the catalytically active, phosphorylated form of both receptors.
- For Western blot analysis, include phosphatase inhibitors in your lysis buffer to preserve phosphorylation. The predicted molecular weights for IGF1R and INSR precursors are ~154 kDa and ~155/156 kDa, respectively; mature receptors may appear as multiple bands due to proteolytic processing.
- When setting up ELISA, consider using a phospho-specific coating method or competition-based format to ensure detection of the activated receptor in cell or tissue lysates.
- Cross-reactivity with mouse and rat has been reported, but validation in your specific sample system is recommended.
