Biological Background: Insulin-like growth factor 1 receptor Function and Localization

  • Insulin-like growth factor 1 receptor (IGF1R), also known as IGF-I receptor, is a receptor tyrosine kinase encoded by the IGF1R gene (GeneID: 3480). It is the primary receptor for insulin-like growth factor 1 (IGF1), binding with high affinity, and also interacts with lower affinity to IGF2 and insulin.
  • Upon ligand binding, IGF1R undergoes autophosphorylation and phosphorylates multiple substrates including IRS1/2, Shc, and 14-3-3 proteins, leading to activation of two major signaling pathways: the PI3K-AKT/PKB pathway and the Ras-MAPK pathway. These pathways promote cell proliferation, inhibit apoptosis, and stimulate protein synthesis, playing a crucial role in cell growth and survival, and are central to tumor transformation.
  • IGF1R can also signal through the JAK/STAT pathway, where phosphorylation of JAK proteins leads to activation of STAT proteins (particularly STAT3), which may be essential for its transforming activity. Additionally, it activates JNK kinases and inhibits JNK activation through phosphorylation and inhibition of MAP3K5/ASK1.

  • Subcellularly, IGF1R is located at the cell membrane. It is widely expressed in tissues including muscle, heart, kidney, adipose tissue, skeletal muscle, hepatoma, fibroblasts, spleen, and placenta, often as hybrid receptors with INSR. It is overexpressed in various tumors like melanomas, and cancers of the colon, pancreas, prostate, and kidney.
  • Post-translational modifications include phosphorylation (critical for its kinase activity and signaling), glycosylation, disulfide bond formation, and cleavage on basic residues. It is also subject to ubiquitination and isopeptide bond formation.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic phosphorylated peptide corresponding to tyrosine 1161 of human IGF1R. This site is a known autophosphorylation site critical for receptor activation. Y1161 phosphorylation is indicative of IGF1R kinase activity. Use this antibody to monitor IGF1R activation status.
  • Suitable for Western blot (WB) and ELISA. In WB, the expected protein weight is approximately 155 kDa (mature, glycosylated receptor) with potential cleaved β-subunit (~95 kDa). Validate with appropriate phosphorylated and non-phosphorylated controls. Cross-reacts with human, mouse, and rat, so the antibody may be useful for studying these species. Consider verification in the relevant sample system.
  • For best results, follow standard protocols for phospho-specific antibodies, including the use of phosphatase inhibitors in lysis buffers. Optimize antibody dilution and detection conditions for your specific experimental setup.
